I used Windows ME DOS Disk as my emergency disk for booting and repairing, i have a Transcend 512 MB USB FLASH Drive. When i boot using my ME Startup disk, i want to be able to access the USB disk like any other hard disk drive (Read/Write). My motherboard supports USB FUNCTION IN DOS. Is This Possible?

Yes, it is possible.
Google for the USB drivers created for FreeDos - maybe them even come included on their full distribution then you only need to adapt them to work with ME (never tested under ME myself, only using Freedos boot disks)

Actually, not to contradict Nuno, just to add some info, the "reference" boot disk should be this one:

if you are using the DUSE drivers.
But usually, say 95% of the times, the Panasonic "Motto Hairu" ones do work.

Actually, I don't really mind being contradicted by a true expert on the matter - also worth mentioning that the more information provided the better!
